Mir-573 regulates cell proliferation and apoptosis by targeting Bax in human degenerative disc cells following hyperbaric oxygen treatment

Abstract Background MicroRNA (miRNA) plays a vital role in the intervertebral disc (IVD) degeneration. The expression level of miR-573 was downregulated whereas Bax upregulated notably human degenerative nucleus pulposus cells. In this study, we aimed to investigate (NP) cells following hyperbaric oxygen (HBO) treatment. Methods NP were separated from degenerated IVD tissues. control maintained 5% CO 2 /95% air and hyperoxic exposed 100% O at 2.5 atmospheres absolute. MiRNA profiling performed via microarray confirmed by real-time PCR, miRNA target genes identified using bioinformatics luciferase reporter assays. mRNA protein levels measured. proliferation NPCs detected MTT assay. Bax, cleaved caspase 9, 3, pro-caspase 3 examined. Results Bioinformatics analysis indicated that 3? untranslated region (UTR) contained “seed-matched-sequence” for hsa-miR-573, which validated MiR-573 induced HBO simultaneous suppression observed Knockdown resulted upregulation HBO-treated addition, overexpression increased cell coupled with inhibition apoptosis. cleavage 9 suppressed while decreased Transfection anti-miR-573 partly effects HBO.
